اثر تیمارهای مختلف بر میزان قهوه‌ای‌شدن و ریز ازدیادی‎ ‎تعدادی از ارقام گیلاس (‏Prunus avium ‎L.‎‏) ‏

چکیده

از آنجایی که گیلاس یکی از مهمترین محصولات باغی مناطق معتدله در کشور محسوب می­شود، استفاده از ریز ازدیادی جهت تسریع در تکثیر آن ضروری است. این مطالعه به منظور بررسی تیمارهای کاهش قهوه­ای شدن و تاثیر نوع و غلظت تنظیم کننده­های رشد بر ریز ازدیادی در قالب آزمایش فاکتوریل بر پایه طرح کاملا تصادفی انجام شد. فاکتور اول، رقم در دو سطح (عدلی و تکدانه)، فاکتور دوم تیمار کاهش قهوه­ای شدن در چهار سطح (تاریکی، سرما، تاریکی و سرما و آنتی اکسیدان (100 میلی­گرم در لیتر اسید آسکوربیک و 150 میلی­گرم در لیتر اسید سیتریک)) و فاکتور سوم تنظیم کننده­های رشد شاخه­زایی شامل BAP (صفر، 1 و 2 میلی­گرم در لیتر) و IBA (صفر و 01/0 میلی­گرم در لیتر) و تیمار NAA (صفر و 1 میلی­گرم در لیتر) و IBA (صفر و 2 میلی­گرم در لیتر) جهت ریشه­زایی مورد مطالعه قرار گرفت. نتایج نشان داد که اختلاف معنی­داری بین تیمارها وجود داشت. بیشترین و کمترین میزان قهوه­ای شدن به ترتیب در تیمار سرما و تیمار آنتی اکسیدان بود. بیشترین تعداد شاخساره در رقم تکدانه و عدلی به ترتیب با میانگین 6/5 و 5 ریز شاخه بود. بالاترین درصد ریشه­زایی در رقم تکدانه 73/78 درصد و در رقم عدلی 4/69 درصد مشاهده گردید. به‌طور کلی، محیط کشت MS حاوی 2 میلی­گرم بر لیتر BAP و 01/0 میلی­گرم در لیتر IBA در مرحله پرآوری و 2 میلی­گرم بر لیتر IBA در مرحله ریشه­زایی برای تکثیر درون شیشه ای رقم عدلی و تکدانه توصیه می­شود.

In Iran, Sweet cherry is one of the most important horticultural products in temperate regions. So it is necessary to use micropropagation methods to accelerate its reproduction. This study was conducted as factorial experiment based on completely randomized design with two cultivars (Adli and Takdaned), and browning reduction treatments in four levels (dark, cold, dark and cold, antioxidant (100 mg/l ascorbic acid and 150 mg/l citric acid)) and proliferation growth regulators BAP at concentrations of (0, 1 and 2 mg/l) and IBA (0 and 0.01mg/l) and rooting stage treatments consisted of NAA (0 and 1 mg/l) and IBA (0 and 2 mg/l).The results showed that there was significant difference between treatments. The highest and lowest levels of browning were in cold treatment and antioxidan treatment, respectively. The highest number of shoots for Takdaneh (5.6) and for Adli (5) cultivars were 5.6 and 5, respectively. The highest rooting percentage was obtained for Takdaneh (78.73%) and Adli (69. 4%). In general, for Adli and Takdaneh cultivars in vitro propagation, MS culture medium containing 2 mg/L BAP and 0.01 mg/L IBA in the proliferation stage and 2 mg/L IBA in the rooting stage is recommended.
