شناسایی، اهلی‌کردن و مقایسۀ عملکرد نژاد بومی قارچ صدفی (Pleurotus spp) با نژاد تجاری

چکیده

در این پژوهش ابتدا دو نژاد قارچ صدفی که از نظر صفات ظاهری تفاوت نشان می‏دادند از جنگل‌های گیلان جمع‌آوری و سپس در محیط کشت MEA کاشته شدند. پس از تکمیل رشد میسیلیوم در ظروف کشت، اسپان قارچ تهیه ‌و کشت قارچ در بستر کاه برنج انجام شد. در‌نهایت پس از برداشت محصول، شناسایی گونه‏های وحشی با استفاده از روش PCR و روش‏های میکرو و ماکروسکوپی صورت گرفت. بر این اساس گونه‏های شناسایی‌شده شامل Pleurotus ostreatus و Pleurotus pulmonariusبودند. نتایج این پژوهش نشان داد که کوتاه‏ترین زمان تشکیل پین‏هد مربوط به نژاد تجاری فلوریدا (8/14روز) و بیشترین میزان پروتئین (24/26 درصد) و نیتروژن (77/9 درصد) مربوط به نژاد استراتوس بود. در سایر صفات اندازه‏گیری شده تفاوت معنا‏داری بین نژادهای مختلف وجود نداشت.

چکیده [English]

In present study two species of Pleurotus with different morphological characteristics were collected from Guilan forest and then cultured on MEA medium. Spawn was prepared on wheat grains and finally all strains were cultivated on rice straw. Identification of species was performed by PCR, microscopic and macroscopic characters. Results revealed that our local strains are known as Pleurotus ostreatus and Pleurotus pulmonarius. Minimum days required for pinhead formation were observed in P. florida (14.8 days). The highest protein and nitrogen content was found in P. florida (26.24 and 9.77%). In other characteristics, there were no any significant differences between strains.
