بررسی تنوع ژنتیکی ژنوتیپ‌های عناب (‏Ziziphus jujuba Mill.‎‏) کلکسیون مرکز تحقیقات و آموزش ‏کشاورزی و منابع طبیعی استان خراسان جنوبی با استفاده از نشانگرهای ‏SSR‏ و ‏RAPD

چکیده

عناب (Ziziphus jujuba Mill.) با کشت طولانی مدت در ایران، دارای ارزش و اهمیت دارویی و اقتصادی می‌باشد. اطلاع از تنوع ژنتیکی جهت مدیریت مؤثر و کارآمد منابع ژنتیکی، شناسایی گونه‌ها و روابط ژنتیکی بین آنها مفید خواهد بود. برای ارزیابی تنوع ژنتیکی 25 درخت عناب کلکسیون مرکز تحقیقات و آموزش کشاورزی و منابع طبیعی استان خراسان جنوبی، از 16 آغازگر RAPD و 22 جفت آغازگر SSR استفاده شد. هر دو نشانگر چندشکلی بالایی نشان دادند. قدرت تمایز نشانگرها با درصد چندشکلی، شاخص محتوای اطلاعات چندشکلی (PIC)، قدرت تفکیک (RP)، شاخص نشانگر (MI) و شاخص مؤثر چندگانه (EMR) مقایسه شد. آغازگرهای RAPD، 108 و در مقابل آغازگرهای SSR، 118 باند چندشکل تکثیر کردند. مقدار متوسط PIC ، EMR و  MIدر نشانگر SSR (به‌ترتیب 47/0، 36/5 و 58/2) نسبت به نشانگر RAPD (به‌ترتیب 19/0، 55/3 و 76/0) بیشتر بود. به‌هرحال نشانگر RAPD مقدار RP بیشتری (92/9) نسبت به SSR (10/2) نشان داد. گروه‌بندی نمونه‌ها براساس دندروگرام ترسیم شده به‌روش UPGMA نشان داد که پاجوش‌های حاصل از یک ژنوتیپ از هم تفکیک شدند که نشان از وقوع جهش سوماتیکی در بین آنها بود. دندروگرام حاصل از ترکیب داده‌های SSR و RAPD، اکثر پاجوش‌ها از ژنوتیپ‌های یکسان را در خوشه‌های یکسانی قرار داد.

Jujube (Ziziphus jujuba Mill.) is cultivated for a long time in Iran, which has medicinal and economic value. In order to effective management of genetic sources, identifying species and determination on the genetic relationships between them, knowledge of genetic diversity will be helpful. To evaluate genetic diversity of 25 trees collected from South Khorasan Agricultural and Natural Resources Research and Education Center Collection, 16 RAPD and 22 SSR primers were used. Both the markers showed highly polymorphic. Distinctive power of two markers were compared using percent of polymorphism, polymorphic information content (PIC), resolving power (RP), marker index (MI) and effective multiplex ratio (EMR). RAPDs produced 108 polymorphic bands, while SSRs amplified 118 polymorphic bands. SSRs showed higher PIC, EMR and MI (0.47, 5.36, 2.58, respectively) than RAPDs (0.19, 3.55, 0.76, respectively). However, RP of RAPDs (9.92) higher than to SSRs (2.10). The dendrogram drawn with UPGMA cluster analysis showed that root-suckers of the same rootstock were separated from each other, which is indicative of the occurrence of somatic mutation among them. Dendrogram derived from the combination of SSR and RAPD datas, divided most of root-suckers from the same rootstock into same clusters.
