Evaluation of callus formation and somatic embryogenesis of Gotu Kola as a medicinal plant

Abstract

For evaluation of somatic embryogenesis of Gotu Kola (Centella asiatica), an experiment was performed in 2019 in tissue culture laboratory of Faculty of Agriculture, University of Guilan. For this purpose, leaf explants were used to induce embryonic calli on MS solid media with 0, 0.25, 0.5, 1.5 and 2 mg/L 2,4-Dichlorophenoxyacetic acid (2,4-D) alone or in combination with 0, 0.25,0.5 and 1 mg/L Benzyl Adenine (BA) growth regulator. The results showed that the 2 mg/L 2,4-D treatment had the highest percentage of callus formation (96.61%), fresh weight (3.96g) and mean callus diameter (3.56cm) in dark conditions. Also calli consisting of 2,4-D treatment at concentrations of 0, 0.1, 0.2 and 0.3 mg/L were used to induction of somatic embryos. The results showed that 0.1 mg/L 2,4-D treatment had the highest mean number of embryos formed at different stages of spherical, cardiac and dragon form. Also, the 0.1 mg/L 2,4-D treatment had the highest percentage of embryogenesis (55.27%) which was not significantly different from 0.2 mg/L 2,4-D treatment. BA growth regulator was used at concentrations of 0, 0.25, 0.5 and 1 mg/L BA to germinate the formed embryos. The results showed that maximum percentage of embryos germination (56.8%) and maximum mean number of seedling production (12.4%) were on 1 mg/L BA treatment.
