تولید و ارزیابی‎ ‎قدرت‎ ‎ترکیب‌پذیری‎ ‎عمومی‎ ‎لاین‌های‎ ‎دابل‌ هاپلویید فلفل دلمه‌ای ‏‏(‏Capsicum annuum L.‎‏)‏

چکیده

ایجاد فناوری‌های کارآمد نظیر دابل هاپلوییدی یک راه حل سریع برای تهیه لاین‌های خالص می باشد. در این تحقیق با به کارگیری روش جنین‌زایی میکروسپور لاین‌های دابل ‌هاپلوئید (اینبرد) فلفل تولید گردید. آزمون تاپ‌کراس با استفاده از 32 لاین دابل هاپلوئید تولیدی و والد تاپ‌کراس California Wonder جهت بررسی ترکیب‌پذیری عمومی لاین‌ها صورت گرفت. هیبریدهای حاصل از تلاقی لاین× ‌تستر در سال زراعی 1397-1398 کشت شدند. به منظور ارزیابی قدرت ترکیب‌پذیری عمومی لاین‌ها صفات روز تا جوانه‌زنی، روز تا گلدهی، روز تا میوه‌دهی، وزن تر میوه، وزن خشک میوه، طول میوه، عرض میوه، قطر گوشت میوه، تعداد بذر در میوه، میانگین عملکرد تک بوته، تعداد میوه در بوته و ارتفاع بوته اندازه‌گیری و تجزیه و تحلیل شدند. مقایسه میانگین‌ها توسط آزمون دانکن نشان داد نتاج تاپ‌کراس لاین‌های دابل هاپلوئید DH53, DH57, DH64, DH55  DH217, DH202,و DH90 به ترتیب با میانگین 58/8، 22/7، 18/7، 99/5، 93/5، 80/5 و 69/5 کیلوگرم در بوته بالاترین میزان عملکرد میوه را داشتند که در این میان بالاترین مقدار مربوط به نتاج تاپ‌کراس حاصل از تلاقی DH55 × California Wonder با میانگین 58/8 کیلوگرم در بوته بود. وراثت‌پذیری صفات مورد ارزیابی بالای 70 درصد بود که در این میان صفات تعداد روز تا جوانه‌زنی و میانگین عملکرد میوه در بوته بیشترین میزان را به خود اختصاص دادند. همچنین لاین‌های اشاره شده بالاترین میزان ترکیب‌پذیری عمومی مثبت و معنی دار برای صفت متوسط عملکرد در بوته را نیز به خود اختصاص دادند. بنابراین می‌توان از این 7 لاین به عنوان لاین‌های والدینی برتر در برنامه‌های اصلاحی آینده استفاده نمود.

Creating new efficient technologies such as doubled haploids is a fast solution for producing inbred lines. In this study, using the microspore embryogenesis method, doubled haploid (inbred) pepper lines were produced. The Topcross test was performed using 32 doubled-haploid lines produced and the topcross parent of California Wonder to examine the general combining ability of the lines. Hybrids derived from crosses line × tester in 2018- 2019 were planted. In order to evaluate the general combining ability of the lines, days to germination, days to flowering, days to fruit set, fresh fruit weight, dry fruit weight, fruit length, fruit width, thickness of fruit diameter, number of seeds, average yield per plant, number of fruits per plant and plant height were measured and analyzed. Comparison of means by Duncan test showed that top-cross progeny of DH55, DH64, DH57, DH53, DH217, DH202, and DH90 doubled haploid lines respectively with average of 8.58, 7.22, 7.18, 5.99, 5.93, 5.80 and 5.69 kg per plant had the highest fruit yield, among them the highest amount was related to the Topcross offset from DH55× California Wonder with an average of 8.58 kg per plant. The heritability of the evaluated traits was over 70%, among them the number of days to germination and the average fruit yield per plant were the highest. The above-mentioned lines also had the highest positive and significant general combining ability for the average yield per plant. Therefore, these 7 lines can be used as superior parent lines in future breeding programs.
