اثر محیط کشت و تنظیم‌کننده‌های رشد گیاهی بر ریزازدیادی پایه نیمه‌پاکوتاه‌کننده گیلاس CAB-6P

چکیده

پایه CAB-6Pبه­عنوان پایه­ای نیمه­پاکوتاهبرایدرختانگیلاس و آلبالو استفاده می­شود.هدف از این تحقیق، تعیین مناسب­ترین محیط ­کشت و تنظیم­کننده­های رشد جهت ریزازدیادی پایه CAB-6P می­باشد. در مرحله پرآوری ریزنمونه­ها در 3 محیط­ کشت MS، DKW و WPM با غلظت­های مختلف تنظیم­کننده رشد BA (5/0، 1 و 2 میلی­گرم در لیتر) در ترکیب با کینتین (صفر و 5/0 میلی­گرم در لیتر) کشت شدند. برای ریشه­زایی از دو محیط­ کشت MS و WPM با 4 غلظت IBA (0، 5/0، 1 و 2 میلی­گرم در لیتر) استفاده شد. نتایج آزمایش نشان داد که محیط کشت WPM حاوی 5/0 میلی­گرم در لیتر BA، با نرخ تکثیر 1/8 شاخساره به­ازای ریزنمونه، مناسب­ترین تیمار پرآوری می­باشد. بالاترین درصد ریشه­زایی به­میزان 69 درصد در محیط کشت MS حاوی 2 میلی­گرم در لیتر IBA به­دست آمد. گیاهچه­های ریشه­دارشده، به بستر کشت پیت‌ماس:کوکوپیت:پرلایت (1:2:2 حجمی) منتقل و 88 درصد گیاهچه­ها با موفقیت سازگار شدند.

چکیده [English]

CAB-6P is used as a semi dwarf rootstock for sweet and sour cherries. The purpose of this study was to determine the best media culture and plant growth regulators on micropropagation of CAB-6P rootstock. Shoot-tips were cultured on three culture media (MS, DKW and WPM) containing different concentrations of BAP (0.5, 1 and 2 mg/L) in combination with Kin (0 and 0.5 mg/L) for proliferation. Multiplied shoots were rooted on MS or WPM media supplemented with IBA (0, 0.5, 1 and 2 mg/L). Results showed that the most suitable multiplication medium was WPM+0.5 mg/L BA with 8.1 shoots per explant. The highest rooting percentage (69%) was obtained with MS+2 mg/L IBA. Well rooted plantlets were transferred to peat moss:cocopeate:perlite (2:2:1 v/v) substrate and successfully hardened (88%).
