اثر سالیسیلیک‌اسید و متیل‌جاسمونات بر تولید هایپریسین در کشت کالوس گل راعی (Hypericum perforatum L.) رقم Topas

چکیده

در این تحقیق اثر سالیسیلیک­اسید و متیل­جازمونات بر تولید هایپریسین در کشت کالوس گلراعی (Hypericum  perforatum L.)  رقمTopas  بررسی گردید. این آزمایش به­صورت فاکتوریل در قالب طرح کاملاً تصادفی با دو فاکتور شامل سالیسیلیک­اسید در چهار سطح (225،150،75،0 میکرومولار) و متیل­جاسمونات در سه سطح (150،75،0 میکرومولار) با سه تکرار در آزمایشگاه گروه پژوهشی علوم­باغبانی دانشگاه علوم­کشاورزی و منابع­طبیعی ساری انجام گرفت. فاکتور­های اندازه­گیری­شده شامل هایپریسین، فنل کل، فلاونوئید کل، آنتوسیانین کل و فعالیت آنزیم فنیل­آلانین­آمونیالیاز (PAL) بود. در مقایسه میانگین اثرات ساده تیمارها، اثر متیل­جازمونات بر فلاونوئید کل معنی­دار شد. اثر متقابل تیمارها برای هایپریسین، فلاونوئید­ها و آنتوسیانین­ها معنی­دار شد.در مجموع، در میان نتایج به‌دست‌آمده از این پژوهش، بیشترین میزان هایپریسین تولید­شده از اثر 150 میکرومولار سالیسیلیک­اسید و همین­طور اثر متقابل 225 میکرومولار سالیسیلیک­اسید (SA) در ترکیب با 75 میکرومولار متیل­جاسمونات (MJ) در مدت زمان 21 روز (سه هفته) به­دست آمد.

چکیده [English]

Effects of salicylic acid and methyl Jasmonate on Hypericin production in callus culture of Hypericum perforatum L. cv Topas were investigated. This factorial experiment was carried out in a completely randomized design with two factors including the four levels of salicylic acid (225,150,75,0 µM) and methyl Jasmonate in three levels (150,75,0 µM) with three replications at the research lab Group of Sari University of Agricultural Sciences and natural resources, deprtment of Horticultural Science. Analysis of variance showed significant differences among treatments. Effect of treatments on total flavonoid Methyl Jasmonate effect was significant. Interaction effect was significant for Hypericin, flavonoids and anthocyanins. According to the results of this study in cell cultures of H. perforatum, low concentration (100 micromolar and lower) of both salicylic acid and methyl Jasmonate elicitors were effective for production of Hypericin. The results of this experiment showed that the highest amount of Hypericin produced from 150 μM salicylic acid (SA) and 225 μM salicylic acid in combination with 75 μM methyl Jasmonate (MJ) after 21 days.
