بررسی چندشکلی نشانگرهای مولکولی SSR و ISSR در نژادگانهای مختلف لیموترش بدون علائم بیماری جاروک
لایمها به دلیل کیفیت و اسیدیتۀ بالای عصارۀ میوه، بهصورت تازه خوری و فرآوری شده استفاده میشود. در سالهای اخیر، رخداد بیماری جاروک، باعث بین رفتن هزاران اصله درخت لایم در جنوب ایران شده است. مهمترین روش کنترل این بیماری، کاشت نژادگان (ژنوتیپ)های متحمل است. بنابراین، در این پژوهش، تنوع ژنتیکی 33 نژادگان مختلف لایم بدون علائم بیماری جاروک در مزرعه یا پس از آزمایشهای مقدماتی، بههمراه ده رقم تجاری مرکبات بهعنوان شاهد با 17 جفت آغازگر SSR و 11 آغازگر ISSR بررسی شد. با توجه به محتوای اطلاعات چندشکلی (PIC) نشانگرهای ریزماهواره TAA1 و TC26 به ترتیب به میزان 72/0 و 68/0 و همچنین در نشانگرهای ISSR، آغازگرهای ISSR1، 809 و 810 هر سه به میزان 49/0 بهعنوان مناسبترین نشانگرهای متمایزکننده شناسایی شدند. نتایج نشان داد، بین لایمها و دیگر انواع مرکبات از لحاظ نشانگرهای مورد بررسی تفاوتهایی وجود دارد. از سوی دیگر همۀ نژادگانهای لایم بر پایۀ هر دو نشانگر، قرابت بالایی با رقمهای مکزیکن لایم و پرشین لایم نشان دادند و در یک خوشه قرار گرفتند. بنابراین، بیشتر نژادگانهای لیموترش در جنوب کشور متعلق به گروه لایمها هستند.

تأثیر برخی از شرایط اقلیمی و خاکی بر کمیت و کیفیت اسانس اکوتیپهای مختلف آویشن دنایی (Thymus daenensis Celak subsp. daenensis)
آویشن دنایی (Thymus daenensis Celak subsp. daenensis) گیاهی است دارویی، اندمیک ایران و متعلق به خانواده نعناع که از میزان اسانس و تیمول اسانس بالایی برخوردار است. ارزیابی اکوتیپهای مختلف این گیاه بهمنظور شناسایی برترین اکوتیپ، از لحاظ بالاترین درصد اسانس و تیمول و نیز تعیین ارتباط خصوصیات خاکی و اقلیمی با کمیت و کیفیت اسانس آن بهمنظور شناسایی مناسبترین اقلیم جهت تعیین محیط کشت ثانویه این گیاه جهت اهلیسازی امری ضروری بهنظر میرسد. در این پژوهش، هشت منطقه (ملایر 1، ملایر 2، جوزان، اراک، خانه میران بالا، خانه میران پایین، شازند، زاغه) از مناطق پراکنش این گیاه انتخاب و مورد مطالعه قرار گرفت. اسانس نمونههای گیاهی جمعآوریشده از هر منطقه استخراج و پس از تعیین بازده اسانس، اجزای اسانس و مقادیر آنها به کمک دستگاههای GC و GC/MS مشخص گردید. ارتباط بین خصوصیات اقلیمی و خاکی مناطق مورد مطالعه با میزان اسانس و اجزای آن مورد بررسی قرار گرفت. نتایج بیانگر آن بود که اکوتیپها اختلاف معنیداری از لحاظ درصد اسانس با یکدیگر داشتند. بیشترین و کمترین درصد اسانس بهترتیب مربوط به اکوتیپهای اراک (5%) و ملایر 2 (5/2%) بود. جزو غالب اسانس همه اکوتیپها، تیمول بود و بیشترین و کمترین درصد تیمول بهترتیب در اسانس اکوتیپهای جوزان (88/78%) و ملایر 1 (16/67%) مشاهده گردید. بین درصد اسانس با میانگین دمای روزانه، رس، سیلت و میزان آهک خاک همبستگی مثبت بالایی وجود داشت که این همبستگیها میتوانند در تعیین محیط کشت ثانویه این گیاه جهت اهلیسازی مفید واقع شوند.

بررسی اثر متقابل نوع محیط کشت و میزان تنظیمکننده رشد در ریزازدیادی پایه پررشد گلابی (Pyrus betulifolia)
بهمنظور دستیافتن به حداکثر پرآوری در محیطهای کشت، میتوان با توجه به نوع گونه گیاهی، از مقادیر مختلف نمکها و تنظیمکنندههای رشد استفاده نمود. در برخی مواقع اثر متقابل این مواد میتواند نتایج متفاوتی را بر میزان پرآوری گیاهچهها، در شرایط درونشیشهای در پی داشته باشد. در این پژوهش تأثیر کاربرد دو محیط کشت پایه MS و QL در ریزازدیادی گلابی گونه Pyrus betulifolia مورد مطالعه قرار گرفت که بیشترین میزان پرآوری در محیط کشت QL و بیشترین میزان توسعهیافتگی برگها در محیط کشت MS بود. بهمنظور تولید برگهای توسعهیافتهتر در محیط کشت QL و با توجه به تفاوت در میزان خالص یونهای NO3-،NH4+، Cl- و Ca2+ در دو محیط کشت مورد بررسی، نمکهای NH4NO3 در سه غلظت 25/6، 5/12 و 75/18 میلیمولار و CaCl2 در دو غلظت 9/0 و 8/1 میلیمولار به محیط کشت QL اضافه شدند. بر اساس نتایج بهدست آمده افزایش در میزان NH4NO3 و CaCl2 تأثیری در میزان پرآوری ریزنمونهها نداشت؛ اما با افزایش در میزان این دو نمک، سطح برگ ریزنمونهها در محیط کشت QL توسعه یافت. در آزمایشی دیگر با هدف افزایش میزان پرآوری در محیط کشت MS، با افزایش در میزان سایتوکینین BAP در این محیط کشت از 5/0 به 1، 5/1 و 2 میلیگرم در لیتر، پرآوری در محیط کشت MS افزایش نشان داد. بهنظر میرسد محیط کشت MS بهدلیل دارا بودن غلظت بیشتری از نمکهای ماکرو در مقایسه با محیط کشت QL، نیازمند تنظیمکننده رشد بیشتری بهجهت دستیافتن به پرآوری مناسب میباشد. در مجموع بر مبنای نتایج این پژوهش، در ریزازدیادی گلابی گونه P. betulifolia با توجه به هدف آزمایش، میتوان از هر دو محیط کشت پایه MS و QL بههمراه غلظت مناسبی از سایتوکینینBAP و نمکهای NH4NO3 و CaCl2استفاده نمود.

بررسی مفهومی و کاربردی باغسازی ایرانی به سبک دوره پیش از اسلام
انسان کاشف طبیعت و عامل برقراری پایداری بقا بوده و هست و این نکته را از شعر، ادب و هنرهای تزیینی ایران در ادوار مختلف تاریخ میتوان یافت، که نشانگر احترام و علاقه ایرانیان به طبیعت است. پژوهشها نشان میدهد که باغسازی در ادوار مختلف تاریخ (چه پیش از اسلام و چه پس از اسلام)، شامل تغییرات و اختلافاتی بوده که بهنظر میرسد تا حدی پیرو تغییر آیینهای دینی و فرهنگی هر دوره میباشد. در این پژوهش سعی شده تا به بررسی کلیاتی از ساختار باغ ایرانی در دوره پیش از اسلام (دورههای هخامنشیان و ساسانیان) پرداخته شود تا با ایدهگرفتن از آثار و باغهای موردی برجای مانده از این دو دوره، بخشهایی (با وسعت 2410 مترمربع) از باغ نمونه کشوری دانشگاه تهران و در زمینی به وسعت 40 هکتار به این سبک طراحی شود و بدینوسیله به درک صحیح موضوع و ملموسبودن آن برای کاربران، که قسمت اعظم آن را دانشجویان تشکیل میدهند، کمک نماید.

تأثیر غلظت درشتمغذیها بر صفات کمی و کیفی گل شاخهبریده سوسن (Lilium LA Hybrid Fangio) در شرایط کشت بدون خاک
این آزمایش به منظور بررسی اثر چهار غلظت از درشت مغذیها بر رشد و عملکرد صفات کمی و کیفی گل شاخه بریده سوسن (Lilium LA Hybrid Fangio) در کشت بدون خاک و در قالب طرح کاملاً تصادفی با چهار تیمار درشت مغذیها، چهار تکرار و ده گلدان در هر تکرار در گلخانه تحقیقاتی دانشگاه گیلان انجام شد. گیاهان با چهار تیمار درشت مغذی (S1، S2، S3 و S4 به ترتیب بیشترین تا کمترین غلظت درشت مغذیها) بر اساس محلول غذایی کوئیک (تغییر یافته) و با غلظتهای متفاوت نمکهای نیترات پتاسیم، دیپتاسیم هیدروژن فسفات، پتاسیم دیهیدروژن فسفات ، نیترات کلسیم، نیترات آمونیوم، کلرید کلسیم و سولفات منیزیم میلیگرم در لیتر محلول آبیاری تغذیه شدند. تمامی محلولهای غذایی حاوی مقادیر مشابهی از عناصر معدنی میکرو بودند. نتایج دادهها نشان داد بین تیمارها از نظر عرض برگ، وزن پیاز دختری، کلروفیل کل برگ، آنتوسیانین گلبرگ، پروتئین گلبرگ و مقدار عناصر نیتروژن، پتاسیم، فسفر و منیزیم موجود در برگ در سطح احتمال 1 درصد و طول غنچه گل، طول برگ و تعداد برگ در سطح احتمال 5 درصد اختلاف معنیداری وجود داشت. درحالیکه در صفاتی مانند ارتفاع گیاه، تعداد گل، تعداد روز تا گلدهی، تولید تعداد پیازچههای دختری، مقدار پرولین، نشت یونی، مقدار آب نسبی برگ و پراکسیده شدن لیپیدها اختلاف، معنیدار نبود.

بررسی ویژگیهای فیتوشیمیایی گل سرخارگل [purpurea (L.) Moench Echinacea] در کشت مخلوط با لوبیا سبز و تاریخهای مختلف کشت تابستانه
در بررسی عملکرد ماده خشک و ویژگیهای فیتوشیمیایی گل سرخارگلهای دوساله در کشت مخلوط سرخارگل با لوبیا سبز و تاریخهای مختلف کشت تابستانه، آزمایشی بهصورت کرتهای خردشده در قالب طرح پایه بلوکهای کامل تصادفی با سه تکرار در سال زراعی 92-1391، طراحی و اجرا شد. تیمارهای آزمایش شامل آرایش کشت [کشت خالص سرخارگل، کشت خالص لوبیا سبز، کشت مخلوط جایگزینی با الگوهای کشت 1:1 (یک در میان) و 2:2 (دو در میان) سرخارگل و لوبیا سبز]، تاریخهای کشت [(10 تیر، 9 مرداد و 8 شهریور) در تابستان 1391] و کشت لوبیا سبز همزمان با سبزشدن سرخارگل در بهار 1392 بود. نتایج آزمایش نشان داد که کشت زودهنگام سرخارگلها در تابستان (10 تیر) موجب افزایش ماده خشک، مشتقات اسید کافئیک و محتوای فنل و فلاونوئید کل گل شد. در میان الگوهای کشت مخلوط نیز، کشت دو در میان سرخارگل و لوبیا سبز، موجب افزایش ماده خشک گل شد. در حالیکه بیشترین مقدار اسید شیکوریک، اسید کلروژنیک و اکیناکوزید بهترتیب با 3/29، 4/2 و 3/2 میلیگرم بر گرم ماده خشک در کشت خالص سرخارگلها بهدست آمد. بیشترین مقدار فنل و فلاونوئید کل گل هم، در تاریخ کاشت 10 تیر و آرایش تککشتی سرخارگلها بهدست آمد. در کل، میتوان گفت که اگر هدف از کشت این گیاه افزایش تولید مشتقات اسید کافئیک باشد، تککشتی و اگر هدف، بهبود تولید ماده خشک باشد، کشت مخلوط مزیت محسوب میشود. به علاوه، کشت زودتر این گیاه نیز از نظر تولید ماده خشک و مشتقات اسید کافئیک نسبت به کشت تأخیری، برتری دارد.

تأثیر سیلیسیوم و سالیسیلیکاسید بر تشکیل لیگنینها و آنزیمهای آنتیاکسیدانت در گل ژربرا
کیفیت و ماندگاری گلها تا حدود زیادی به پایداری ساقه و پتانسیل آنتیاکسیدانی بستگی دارد. این پژوهش برای مطالعه اثر تغذیه سیلیسیوم و تیمار سالیسیلیکاسید بر کیفیت گل ژربرا (Gerbera jamesonii Bolus.) و فعالیت آنزیمهای آنتیاکسیدانت در مراحل پس از برداشت، بهصورت کشت بدون خاک در قالب دو آزمایش فاکتوریل با طرح بلوکهای کامل تصادفی انجام شد. آزمایش اول شامل محلولپاشی قبل از برداشت صفر، 10 و 20 میلیمولار سیلیسیوم (Si) و سالیسیلیکاسید (SA) صفر، 100 و 200 میکرومولار بود. در آزمایش دوم تیمار سیلیکاتپتاسیم همانند آزمایش اول و تیمار سالیسیلیکاسید صفر، 200 و 400 میکرومولار پس از برداشت انجام شد. نتایج نشان داد که طول شاخه گل تحت تأثیر تغذیه سیلیسیوم تاحدودی کاهش یافت (05/0p≤) ولی سیلیسیوم (001/0p≤) و محلولپاشی سالیسیلیکاسید (05/0p≤) موجب افزایش تعداد گل شدند. سالیسیلیکاسید باعث تقویت بیوسنتز لیگنینها (001/0p≤) در ساقه گردید. مصرف سیلیسیوم موجب ماندگاری بیشتر گلها (001/0p≤) گردید ولی افزایش فعالیت آنزیم پراکسیداز و کاتالاز تحت تأثیر تیمار بعد از برداشت سالیسیلیکاسید (SA) (001/0p≤) بود. این نتایج نشان داد که تغذیه سیلیسیوم و کاربرد سالیسیلیکاسید میتواند کیفیت و ماندگاری گل بریده ژربرا را بهبود بخشد.

تأثیر اسیدسالیسیلیک و متیلجاسمونات بر ویژگیهای مورفولوژیکی، فعالیت آنزیمیهای آنتیاکسیدانی و درصد اسانس گیاه سرخارگل (Echinacea purpurea L.) در شرایط اقلیمی زنجان
بهمنظور ارزیابی اثر اسیدسالیسیلیک و متیلجاسمونات و اثرات متقابل آنها بر برخی از ویژگیهای گیاه سرخارگل، آزمایشی بهصورت فاکتوریل بر پایه طرح بلوکهای کامل تصادفی در سه تکرار، در شرایط اقلیمی زنجان اجرا شد. تیمارهای آزمایش شامل سه سطح اسیدسالیسیلیک (صفر، 1 و 2 میلیمولار) بهعنوان فاکتور اول و متیلجاسمونات در سه سطح (صفر، 5/0 و 1 میلیمولار) بهعنوان فاکتور دوم بودند. صفات مورد ارزیابی شامل صفات مورفولوژیکی (ارتفاع بوته، سطح برگ، تعداد گل، درصد وزن خشک بوته)، صفات فیزیولوژیکی (کلروفیل کل، کاروتنوئید، فلاونوئید) و فعالیت آنزیمهای آنتیاکسیدانی پراکسیداز و کاتالاز و همچنین درصد اسانس بودند. بر پایه نتایج بهدستآمده در این پژوهش، اثر متقابل 1 میلیمولار اسیدسالیسیلیک و 5/0 میلیمولار متیلجاسمونات بیشترین اثر را بر فعالیت آنزیمهای کاتالاز و پراکسیداز داشت. کاربرد برگی اسیدسالیسیلیک 2 میلیمولار باعث افزایش ارتفاع، سطح برگ و تعداد گل شد. در بین غلظتهای مختلف متیلجاسمونات، غلظت 1 میلیمولار این ماده بیشترین اثر را بر مقدار کلروفیل کل، کاروتنوئید و فلاونوئید در مقایسه با تیمارهای اسیدسالیسیلیک داشت و غلظت 2 میلیمولار اسیدسالیسیلیک، بیشترین اثر را بر مقدار درصد اسانس نسبت به تیمارهای متیلجاسمونات بر جای گذاشت. با توجه به نتایج این پژوهش، در مجموع اثر ساده کاربرد برگی 2 میلیمولار اسیدسالیسیلیک و 1 میلیمولار متیلجاسمونات توانست اکثر صفات اندازهگیریشده در این پژوهش را نسبت به تیمار شاهد بهطور معنیداری بهبود بخشد.

تعیین ارزش غذایی و ظرفیت پاداکسایشی اندامهای مختلف دو گونه والک ایرانی (Allium akaka S.G. Gmelin و Allium elburzense W.) در شرایط اقلیمی رویشگاههای مختلف و مزرعه
والک یکی از سبزیهای بومی ایران است که چندین گونه از جنس آلیوم و زیرجنس ملانوکرومیوم را شامل میشود. این تحقیق با هدف بررسی ارزش غذایی و ظرفیت پاداکسایشی اندامهای مختلف هشت جمعیت وحشی از دو گونه والک ایرانی (Allium akaka S.G. Gmelin و Allium elburzense W.) در اقلیمهای مختلف انجام شد. نتایج نشان داد مقدار پتاسیم و کلسیم در رویشگاه اصلی مورد مطالعه، بهترتیب بهمیزان 182 تا 432 و 159 تا 252 (mg/100g) و در مزرعه نیز بهترتیب 311 تا 533 و 193 تا 243 (mg/100g) بهدست آمد. مقدار فنل کل بهمیزان 73/1 تا 57/2 (g AGE/kg FW)، مقدار اسید آسکوربیک بهمیزان 08/6 تا 11/8 (mg/100g FW) و ظرفیت پاداکسایشی در بازه 13/2 تا 02/3 (g AAE/kg FW) در مزرعه بهدست آمد؛ درحالیکه میزان این ترکیبات در رویشگاه اصلی بهترتیب در بازه 95/1 تا 52/2(g AGE/kg FW)، 15/6 تا 94/8 (mg/100g FW) و 85/1 تا 07/3 (g AAE/kg FW) بود. ضرایب همبستگی کانونیک نشان دادند که با افزایش ارتفاع از سطح دریا، pH خاک و کاهش میانگین دما، مقدار عناصر روی، آهن و پتاسیم افزایش یافت، که منجر به افزایش ترکیبات دفاعی گیاه، از جمله فنلها و در نتیجه ظرفیت پاداکسایشی کل شد. همچنین نتایج نشان داد کلسیم، پتاسیم، منیزیم و روی بیشترین اثر را بر سنتز اسید آسکوربیک، فنلها و ظرفیت پاداکسایشی، نسبت به سایر عناصر داشتند. بر اساس نتایج بارهای کانونیک، تمام فراسنجههای محیطی بر ارزش غذایی و ظرفیت پادکسایشی بافت والک اثرگذار بودند که بررسی هرکدام از آنها بهتنهایی و یا چند مورد از آنها، منجر به نتیجه مطلوب نمیگردد و تمام این عوامل در کنار یکدیگر، شرایط موجود گیاه را توجیه میکنند.

تأثیر نانونقره بر آلودگی باکتریایی و شاخصهای مورفولوژیکی و بیوشیمیایی بادام GN15 کشت بافتی
نانوتکنولوژی توانسته راهگشای روشهای نوینی در علوم گیاهی و تحقیقات کشاورزی باشد. این تحقیق بهمنظور بررسی اثرات نانونقره در 4 غلظت (صفر (شاهد)، 100، 150 و 200 میلیگرم بر لیتر) و 8 تکرار، در قالب طرح بلوکهای کامل تصادفی، بر درصد زندهمانی، آلودگی باکتریایی و شاخصهای مورفولوژیکی و بیوشیمیایی ریزنمونههای بادام GN15، در دانشگاه ارومیه، اجرا شد. جوانه انتهایی بادام رشدیافته در محیط کشت MS حاوی 3 درصد ساکاروز و 8/0 درصد آگار، در محیط مشابه، حاوی غلظتهای مختلف نانونقره و 1 میلیگرم بر لیتر BAP، واکشت و در اتاقک رشد قرار داده شدند. با افزایش غلظت نانونقره، درصد زندهمانی ریزنمونهها افزایش و آلودگی باکتریایی و قارچی کاهش یافتند. نانونقره در غلظتهای صفر تا 100 میلیگرم بر لیتر، باعث افزایش طول ریشه، تعداد برگ، میزان کلروفیل a، b، کلروفیل کل، کارتنوئید و کربوهیدراتهای محلول برگ و در غلظتهای بالاتر، موجب کاهش این فاکتورها شد. افزایش غلظت نانونقره باعث کاهش طول ساقه و تعداد ساقه شد، اما روی تعداد ریشه تأثیر معنیداری نداشت. با وجود تأثیر افزایش غلظت نانونقره در کاهش آلودگی باکتریایی، تا سطح 200 میلیگرم بر لیتر، حتی تا کمتر از 10 درصد، بهترین سطح، درجهت بهبود ویژگیهای رویشی و بیوشیمیایی ریزنمونههای GN15، تیمار 100 میلیگرم بر لیتر بود.

مطالعه تنوع ژنتیکی برخی لاینهای فلفل (Capsicum annuum L.) با استفاده از تجزیههای آماری چند متغیره
بهمنظور بررسی تنوع ژنتیکی 65 لاین فلفل، که از مناطق مختلف کشور و همچنین از کشور چین و مرکز تحقیقات بینالمللی سبزیجات (AVRDC) جمعآوری شده بودند؛ آزمایشی بهصورت طرح بلوکهای کامل تصادفی با سه تکرار برای ارزیابی 13 صفت مرفولوژیک اجرا شد. نتایج حاصل از تجزیه واریانس حاکی از تفاوت معنیدار بین ژنوتیپها در سطح احتمال یک درصد از لحاظ تمامی صفات بود. بر اساس مقایسه میانگین صفات مختلف که بهروش توکی در سطح احتمال 5 درصد انجام شد، ژنوتیپهای 3095 و 0013 و 26-6 از لحاظ عملکرد و خصوصیات مرتبط با بازارپسندی محصول وضعیت بهتری داشتند. برای تعیین فاصله ژنتیکی و گروهبندی ژنوتیپها، تجزیه به عاملها و تجزیه خوشهای انجام شد. جهت درک بهتر از روابط داخلی صفات و تعیین گروهی متغیرهای با بیشترین همبستگی، از تجزیه به عاملها با استفاده از تجزیه به مؤلفههای اصلی و چرخش عاملها به روش وریماکس استفاده شد. در این تجزیه چهار عامل مستقل از هم، مجموعاً 07/74 درصد از تغییرات را توجیه نمودند. تجزیه خوشهای بهروش وارد، لاینهای ارزیابیشده را در شش گروه متفاوت قرار داد؛ بهطوریکه این نتایج تا حد زیادی با مناطق جغرافیایی پراکنش لاینها و همچنین با صفاتی همچون عملکرد و خصوصیات مورفولوژیک مرتبط با بازارپسندی محصول همخوانی داشتند. نتایج حاصل از رگرسیون گامبهگام نشان داد که سه صفت قطر ساقه، طول ساقه و تعداد میوه در بوته بیشترین تأثیر را بر روی عملکرد میوه داشتند. جهت ارزیابی اثرات مستقیم و غیرمستقیم صفات واردشده در مدل رگرسیونی بر عملکرد میوه، تجزیه علیت انجام شد. نتیجه حاصل از این تجزیه موید تأثیر مستقیم سه صفت یادشده بر روی عملکرد بود و تأثیر غیرمستقیم این صفات چندان قابلتوجه نبود. با توجه به اینکه در این آزمایش لاینهای مورد مطالعه از لحاظ تنوع ژنتیکی تفاوتهای قابلتوجهی در صفات مورد بررسی نشان دادند نتیجهگیری شد که با انتخاب ژنوتیپهای برتر در هر خوشه که فاصله ژنتیکی بیشتری با یکدیگر دارند میتوان در تولید ارقام هیبرید فلفل بهمنظور بهرهجستن از هتروزیس استفاده کرد.

ماندگاری و واکنش فیزیولوژیکی گل شاخهبریده لیلیوم اورینتال به عنصر شبهضروری سیلیسیم در کشت بدون خاک
این آزمایش با توجه به حذف خاک در کشتهای بدون خاک و در نتیجه حذف منبع اصلی تأمین عنصر شبهضروری سیلیسیم و همچنین عدم مطالعه تأثیر احتمالی این عنصر در پرورش لیلیوم شاخهبریده، طراحی و اجرا گردید. در همین راستا و پس از انتقال پیازهای لیلیوم (Lilium oriental 'Casa Blanca') به بستر فیبر نارگیل (الیاف فرآوریشده نارگیل)، گیاهان استقرار یافته با محلول غذایی حاوی غلظتهای مختلف اسیدسیلیسیک (صفر، 50، 100 و 150 میلیگرم در لیتر سیلیسیم) محلولدهی و ویژگیهای فیزیولوژیکی و کیفیت پس از برداشت گلهای تولیدی در قالب طرح کاملاً تصادفی با 3 تکرار مورد بررسی قرار گرفت. نتایج آزمایش بیانگر افزایش معنیدار آب جذبشده توسط ساقه گلدهنده، ماندگاری گل، کلروفیل کل، قند محلول، محتوای نسبی آب و کاهش معنیدار نشت یونی برگ بود. بر همین اساس، بیشترین آب جذبشده توسط ساقه گلدهنده (6/0 میلیلیتر بر گرم وزنتر در روز)، عمر گلجایی (11/11 روز) و کلروفیل کل (97/0 میلیگرم بر گرم وزنتر) در غلظت 150 میلیگرم در لیتر سیلیسیم مشاهده گردید. همچنین جذب عناصر معدنی پتاسیم، کلسیم و سیلیسیم بهطور معنیداری تحت تأثیر سیلیسیم قرار گرفتند. بیشترین غلظت کلسیم درغلظت 150 میلیگرم در لیتر سیلیسیم مشاهده شد. براساس نتایج آزمایش، افزودن سیلیسیم به محلول غذایی منجر به بهبود روابط آبی گیاه، شاخصهای کیفی پس از برداشت، جذب عناصر معدنی و در نتیجه افزایش ماندگاری گل شاخهبریده لیلیوم رقم کاسابلانکا گردید.

شناخت و ارزیابی صفات کیفی میوه هلو از نظر برنامههای بهنژادی و حفظ ژرمپلاسم بسیار با اهمیت است. برای این منظور، بررسی و مقایسه خواص کیفی و بیوشیمیایی میوه 18 ژنوتیپ و رقم هلوی موجود در استان آذربایجانشرقی در قالب طرح بلوکهای کامل تصادفی با سه تکرار انجام گرفت. صفات مختلفی از قبیل سفتی بافت میوه، مواد جامد محلول (TSS)، pH آب میوه، اسیدیته قابلتیتراسیون (TA)، ویتامین ث و محتوای فنل کل و ظرفیت آنتیاکسیدانی، فلاونوئید کل و آنتوسیانین کل در دو بخش پوست و گوشت مورد ارزیابی قرار گرفت. همچنین بهمنظور تعیین ارتباط بین صفات، همبستگی ساده بین صفات نیز محاسبه گردید. نتایج حاصله نشان داد که اثر ژنوتیپ و رقم روی بیشتر صفات مورد مطالعه در سطح احتمال 1 درصد معنیدار بود. صفات بیوشیمیایی مورد بررسی در همه ارقام و ژنوتیپهای ارزیابیشده در بخش پوست بیشتر از گوشت بود. همچنین نتایج نشان داد که رقم هلوی حامدی دارای بیشترین مقدار آنتوسیانین کل و فلاونوئید کل بود و ارقامی مانند انجیری موری، انجیری زعفرانی خلیلی دارای بیشترین محتوی ویتامین ث بوده، ظرفیت آنتیاکسیدانی و محتوای فنل کل بودند. نتایج همبستگی نشان داد که محتوی ویتامین ث با ظرفیت آنتیاکسیدانی، فنل کل و فلاونوئید کل گوشت و پوست همبستگی معنیداری نداشت، درحالیکه همبستگی مثبت و معنیداری بین فنل کل و ظرفیت آنتیاکسیدانی مشاهده گردید. بهطور کلی با توجه به نتایج بهدست آمده ارقام با ترکیبات بیوشیمیایی و آنتیاکسیدانی بالا میتواند در برنامههای اصلاحی مورد استفاده قرار گیرند.

اثر اسانسهای مرزه و میخک روی عمر گلجایی، تعادل آبی شاخه و برگ در گل شاخه بریده رز رقم ولوت
این آزمایش بهمنظور بررسی اثر اسانسهای گیاهی مرزه (Satureja hortensis) و میخک(Syzygium aromaticum) در غلظتهای صفر، 250 و 500 میلیگرم در لیتر روی عمر گلجایی گل شاخه بریده رز رقم ولوت انجام شد. آزمایش تحت شرایط دمای 2±20 درجه سانتیگراد، رطوبت نسبی 70-60 درصد، شدت نور سفید 12 میکرومول در متر مربع در ثانیه و 12 ساعت طول روز انجام شد. این آزمایش بهصورت فاکتوریل و در قالب طرح کاملاً تصادفی با 18 تیمار با 3 تکرار و 3 گل شاخه بریده در هر گلدان برای اندازهگیری صفات عمر گلجایی، وزن تر نسبی شاخه، جذب محلول شاخه، هدایت روزنهای برگ، نسبت فلورسنس کلروفیل و محتوی کلروفیل برگ در هر تکرار صورت گرفت. تیمار 500 میلیگرم در لیتر از اسانسهای گیاهی منجر به حفظ عمر گلجایی شاخه بریدهها شد. 250 میلیگرم در لیتر از اسانسها شاخص کلروفیل برگ و جذب محلول را در مقایسه با سایر غلظتها افزایش داد. تیمار محلول ساکارز 2 درصد توانست باعث حفظ معنیدار عمر گلجایی، نسبت فلورسانس کلروفیل و محتوی کلروفیل برگ نسبت به تیمار ساکارز صفر شود. اسانسهای مرزه و میخک بهترتیب در غلظتهای 500 و 250 میلیگرم در لیتر باعث حفظ وزن تر نسبی شاخه و کاهش هدایت روزنهای برگ شدند.

اثر زمان برداشت و کیتوزان بر کیفیت و عمر انبارمانی میوههای عروسک پشتپرده (Physalis angulata L.)
رسیدن میوه شامل تغییرات اساسی فیزیولوژی و بیوشیمیایی است که رنگ، طعم، عطر، بافت و ارزش غذایی میوهها را تغییر میدهد. در طول انبارداری و پس از آن، رسیدن کامل میوهها ادامه مییابد، اگر این میوهها مصرف یا فراوری نشوند، بیش از حد رسیده و کیفیت آنها سریعاً روبهزوال میرود. با توجه به اینکه برداشت در زمان مناسب میتواند عمر مفید میوه را بهبود بخشد؛ بهمنظور مطالعه اثر مرحله برداشت و پوشش کیتوزان بر انبارمانی و کیفیت میوههای عروسک پشتپرده، آزمایشی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی در سه تکرار انجام شد. تیمارهای آزمایش شامل مرحله برداشت (سبز بالغ، سبز مایل به زرد و زردرنگ) و پوشش کیتوزان (صفر، 5/0 و 1 درصد) بود. میوههای برداشت و تیمارشده بهمدت 30 روز در دمای 15 درجه سانتیگراد نگهداری شدند. نتایج نشان داد که اثر مرحله برداشت و سطوح مختلف پوشش کیتوزان طی انبارمانی تأثیر معنیداری روی صفات کیفی و عمر انباری میوه داشت. با بلوغ میوه درصد کاهش وزن، میزان ویتامینث، کارتنوئید و شاخص طعم بهطور معنیداری افزایش یافت و میزان کلروفیل، فنل کل و اسیدیته قابلتیتراسیون کاهش یافت. کاربرد پوشش کیتوزان کاهش محتوای کلروفیل، فنل کل، اسیدیته قابلتیتراسیون، درصد کاهش وزن، روند بلوغ و تغییر رنگ میوه را در مدت انبارمانی به تأخیر انداخت. بیشترین میزان کاهش وزن میوه، شاخص طعم، ویتامینث طی بلوغ و انبارداری در میوههای شاهد مشاهده شد. همچنین میوههای سبز بالغ تیمارشده با کیتوزان در سطح 5/0 درصد، بهترین کیفیت طی انبارمانی داشت.

اثر محیط کشت و تنظیمکنندههای رشد گیاهی بر ریزازدیادی پایه نیمهپاکوتاهکننده گیلاس CAB-6P
پایه CAB-6Pبهعنوان پایهای نیمهپاکوتاهبرایدرختانگیلاس و آلبالو استفاده میشود.هدف از این تحقیق، تعیین مناسبترین محیط کشت و تنظیمکنندههای رشد جهت ریزازدیادی پایه CAB-6P میباشد. در مرحله پرآوری ریزنمونهها در 3 محیط کشت MS، DKW و WPM با غلظتهای مختلف تنظیمکننده رشد BA (5/0، 1 و 2 میلیگرم در لیتر) در ترکیب با کینتین (صفر و 5/0 میلیگرم در لیتر) کشت شدند. برای ریشهزایی از دو محیط کشت MS و WPM با 4 غلظت IBA (0، 5/0، 1 و 2 میلیگرم در لیتر) استفاده شد. نتایج آزمایش نشان داد که محیط کشت WPM حاوی 5/0 میلیگرم در لیتر BA، با نرخ تکثیر 1/8 شاخساره بهازای ریزنمونه، مناسبترین تیمار پرآوری میباشد. بالاترین درصد ریشهزایی بهمیزان 69 درصد در محیط کشت MS حاوی 2 میلیگرم در لیتر IBA بهدست آمد. گیاهچههای ریشهدارشده، به بستر کشت پیتماس:کوکوپیت:پرلایت (1:2:2 حجمی) منتقل و 88 درصد گیاهچهها با موفقیت سازگار شدند.

انگور یکی از مهمترین محصولات باغی است که بهدلیل ارزش اقتصادی و غذایی آن بهطور گسترده کشت میشود. بهمنظور دستیابی به ارقام جدید با عملکرد بالا و صفات کیفی بهتر، شناسایی ارقام موجود ضروری است. برخی از گونههای وحشی انگور در معرض فرسایش ژنتیکی قرار دارند؛ لذا تعیین تنوع ژنتیکی ارقام و ژنوتیپهای وحشی انگور بهمنظور توسعه برنامههای اصلاحی آینده انگور دارای اهمیت بالایی میباشد. در این تحقیق تنوع ژنتیکی 75 رقم و ژنوتیپهای وحشی انگور (Vitis vinifera L.) با استفاده از 17 نشانگر ISSR مورد بررسی قرار گرفت. در مجموع 132 باند با استفاده از 17 نشانگر ISSR تولید شد که از این تعداد 75 باند (58/57%) چندشکل بودند. تعداد باندهای چندشکل از 2 در آغازگر UBC873 تا 7 در آغازگز UBC836 متغیر بود. تعداد آللهای مؤثر (Ne) از 72/1 در آغازگر UBC880 تا 18/1 در آغازگر UBC873 متغیر بود. محتوای اطلاعات چندشکل (PIC) آغازگرها بین 42/0 در آغازگر UBC880 و 14/0 در آغازگر UBC873 با میانگین 32/0 برآورد شد. گروهبندی ژنوتیپها بهروش تجزیه خوشهایی بر اساس ماتریس تشابه Dice و با استفاده از الگوریتم Complete ارقام مورد مطالعه را در چهار گروه قرار داد. بیشترین تشابه ژنتیکی (73/0) بین ارقام جیغجیغا و بلکسیدلس و همچنین بین آلفونسو و بلکسیدلس و کمترین تشابه ژنتیکی (11/0) بین ارقام دسترچین و لعلسیاه مشاهده گردید. بر اساس شاخص شباهت ژنتیکی Nei، بیشترین فاصله ژنتیکی مربوط به جمعیتهای خارجی و وحشی بود. آنالیز واریانس مولکولی تنوع درون جمعیتها و بینجمعیتی را بهترتیب 61 درصد و 39 درصد از کل تنوع نشان داد.

اثر پرولین و اسید سالیسیلیک بر شاخصهای مورفولوژیکی و فیتوشیمیایی استویا (Stevia rebaudiana Bert.)
در این مطالعه بهمنظور بررسی اثر پرولین و اسید سالیسیلیک بر صفات مورفولوژیکی و فیتوشیمیایی گیاه استویا آزمایشی بهصورت طرح فاکتوریل در قالب بلوکهای کامل تصادفی با سه تکرار در مزرعه تحقیقاتی دانشگاه زنجان اجرا شد. تیمارهای آزمایش شامل پرولین در سه سطح (صفر، 1، 2 میلیمولار) و اسید سالیسیلیک در سه سطح (صفر، 3/0، 6/0 میلیمولار) بودند که بهصورت محلولپاشی برگی پس از استقرار نشا، در سه مرحله به فاصله 10 روز انجام شد. صفات مورد بررسی شامل سطح برگ، عملکرد وزن تر و خشک برگ و کل بوته در واحد سطح، کلروفیل (a،b و کل)، کاروتنوئید، فنل کل، فلاونوئید کل و درصد فعالیت آنتیاکسیدانی بودند. تیمارها بهطور قابلتوجهی در صفات بالا نسبت به شاهد تأثیرگذار بودند. بالاترین میانگین سطح برگ (cm239/5949)، عملکرد تر (g/m217/787) و خشک برگ (g/m25/228) و عملکرد تر (g/m223/1339) و خشک کل بوته (g/m252/327) در سطح تیمار 1 میلیمولار پرولین بههمراه 3/0 میلیمولار اسید سالیسیلیک مشاهده شد. کاربرد اثرات ساده غلظتهای 2 میلیمولار پرولین و 6/0 میلیمولار اسید سالیسیلیک منجر به افزایش میزان کلروفیل a، b، کاروتنوئید و فلاونوئید کل شد. بالاترین میزان کلروفیل کل (mg/g fw23/5) در اثرات متقابل تیمار 2 میلیمولار پرولین بههمراه 3/0 میلیمولار اسید سالیسیلیک و بالاترین میزان فنل (mg/g dw94/19) در 1 میلیمولار پرولین بههمراه 6/0 میلیمولار اسید سالیسیلیک مشاهده شد. میتوان استنباط نمود که تیمار با پرولین و اسید سالیسیلیک با تأثیر بر فرآیندهای رشدی و فیزیولوژیکی، در افزایش عملکرد و تولید متابولیتهای ثانویه استویا مؤثر بودند.

اثر محلولپاشی زمستانه اوره برگلدهی، عملکرد و تغییرات عناصر معدنی دراندامهای هوایی نارنگی کینو
نیتروژن نقش مهمی در مدیریت تغذیه درخت در مرکبات ایفا میکند. آزمایشی بهمنظور بررسی اثر سطوح مختلف اوره (صفر، 75/0%، 5/1%)، و زمانهای مختلف محلولپاشی اوره (اول دیماه، 15 دیماه، 30 دیماه)، بهصورت فاکتوریل در قالب طرح بلوکهای کامل تصادفی با سه تکرار بر میزان نیتروژن، فسفر، پتاسیم، برگ و گره و خصوصیات گل و عملکرد گیاه نارنگی رقم کینو، صورت گرفت. در این آزمایش از برگ و گره در 1، 3 و 5 هفته بعد از محلولپاشی نمونهبرداری انجام شد. نتایج این پژوهش نشان داد که سه هفته بعد از محلولپاشی 1 دی، بیشترین مقدار نیتروژن برگ (5/2%) مربوط به گیاهان محلولپاشیشده با غلظت 5/1% اوره بود درحالیکه بیشترین مقدار نیتروژن گره (73/1%) در گیاهان تیمارشده در 30 دیماه با غلظت 5/1% اوره بهدست آمد. بیشترین مقدار پتاسیم و فسفر برگ گیاهان محلولپاشی در 30 دیماه با غلظت 5/1% اوره گزارش شد. همچنین غلظتهای مختلف اوره موجب افزایش تعداد گل، تحریک رشد تخمدان و افزایش عملکرد شد و بیشترین تأثیر مربوط به بالاترین غلظت اوره (5/1%) گزارش شد.

مطالعه خصوصیات مورفولوژی و فنولوژی، میزان عناصر غذایی پر مصرف (نیتروژن، فسفر، پتاسیم) و درصد اسانس چهار جمعیت گیاه سنبلهای کرکدار (Stachys pilifera L.)
در این پژوهش پس از شناسایی رویشگاههای گیاه سنبلهایکرکدار (Stachys pilifera L.) در چهار منطقه از استان کهگیلویه و بویراحمد، از هر رویشگاه 10 نمونه کامل گیاهی در زمان گلدهی برای ارزیابی ریختشناسی و سرشاخههای گلدار برای استخراج اسانس جمعآوری شد. مشخصات جغرافیایی و اقلیمی مربوط به هر رویشگاه یادداشتبرداری گردید و از هر رویشگاه یک نمونه خاک، برای ارزیابی خصویات فیزیکی و شیمیایی خاک، نمونهگیری شد. نتایج بهدستآمده با استفاده از نرمافزار آماری SAS تجزیه آماری شد و همبستگی بین صفات ارزیابی گردید. نتایج نشان داد که بیشترین درصد نیتروژن (11/1%)، فسفر (233/0%) و پتاسیم اندام هوایی (46/1%) و بیشترین میزان اسانس (8/0%) مربوط به گیاهان رویشگاه لوداب و کمترین این صفات نیز مربوط به گیاهان رویشگاه سپیدار بود. همچنین نشان داده شد که تمامی صفات مورفولوژیک اندازهگیریشده در رویشگاهها، با افزایش ارتفاع کاهش یافت. در نهایت گیاه سنبلهای کرکدار واقع در رویشگاههای با ارتفاع کمتر از سطح دریا، بهدلیل داشتن خصوصیات رویشی و زایشی بهتر؛ از قبیل بالا بودن طول ساقه گلدهنده، طول و عرض برگ و همچنین عملکرد بالای اسانس، میتواند در برنامههای اصلاحی یا برای اهلیسازی مورد توجه قرار گیرد.
