In this investigation, biochemical composition profiling of the healthy and infected rooted Mexican lime wasmonitored during WBDL progression (90-330 days) as well as to distinguish infected from healthy Mexican limes under greenhouse conditions. For this aim, stem cuttings is taken from healthy Mexican lime. After three months, half of Mexican lime cutting were inoculated by bark-grafting from WBDL-affected Mexican. Six healthy and infected plants were sampled 90, 150, 210, 270 and 330 days after inoculation with Phytoplasma. Pigments, total phenol, antioxidant capacity, soluble carbohydrate and starch content in leaves were measured. Results showed Phytoplasmal infection caused growth inhibition and reduced stem and root dry mater. Moreover, infection mostly caused to a marked decrease in pigments and significantly increased soluble carbohydrates and starch. In Mexican lime leaves, phenol contents and antioxidant capacity were increased after inoculation with Phytoplasma. In addition, the principal components analysis (PCA) revealed a clear distinction between the leaf biochemical profiles of healthy and infected plants during the progression of WBDL. In conclusion, this study provides new insights into the lime response to phytoplasma infection during the progression of WBDL. Finally, we showed that healthy plants can be isolated from infected plants using biochemical profiles.}, keywords = {Candidatus Phytoplasma aurantifolia,PCA,phenol,pigments,starch}, title_fa = {پایش ترکیبات بیوشیمیایی در برگ مکزیکن‌لایم طی پیشرفت بیماری جاروک لیموترش}, abstract_fa = {بیماری جاروک لیموترش، که توسط Candidatus Phytoplasma aurantifolia ایجاد می­شود، مهم‌ترین عامل محدودکننده تولید لیموترش در مناطق جنوبی ایران است. بنابراین تحقیق حاضر با هدف بررسی تغییرات برخی ویژگی‏های بیوشیمیایی در برگ نهال‌های ریشه‌دار مکزیکن‌لایم آلوده‌شده با فیتوپلاسما و مکزیکن‌لایم سالم طی پیشرفت بیماری (330-90 روز) و امکان جداسازی نهال­های سالم و آلوده تحت شرایط گلخانه­ای پایه­ریزی شد. بدین‏منظور، قلمه­هایی از نهال سالم مکزیکن‌لایم تهیه شد. پس از سه ماه، نیمی از قلمه­های مکزیکن‌لایم با استفاده از پیوندک پوست از نهال آلوده به بیماریگر فیتوپلاسما آلوده شدند. در ادامه تعدادی برگ از هر یک از شش نهال سالم و شش نهال آلوده در بازه­های زمانی 90، 150، 210، 270 و 330 روز پس از آلودگی نمونه­برداری شد و مقدار رنگدانه‏ها، فنل کل، ظرفیت آنتی­اکسیدان، کربوهیدرات محلول و نشاسته در این برگ­ها اندازه­گیری شد. نتایج نشان داد که آلودگی با فیتوپلاسما منجر به کاهش چشمگیر رشد و نیز کاهش وزن ماده خشک بخش هوایی و ریشه نهال‏های مکزیکن‌لایم شد. علاوه بر این، آلودگی غالباً منجر به کاهش مقدار رنگدانه­ها و افزایش معنی­دار مقدار کربوهیدرات محلول و نشاسته شد. همچنین، آلودگی با فیتوپلاسما منجر به افزایش معنی­دار فنل کل و ظرفیت آنتی‏اکسیدان در برگ گیاه میزبان شد. به­علاوه، نتایج تجزیه به مؤلفه‌های اصلی نشان داد که براساس شاخص­های بیوشیمیایی اندازه‏گیری­شده در برگ گیاهان سالم و آلوده طی پیشرفت بیماری، می­توان این گیاهان را از یکدیگر تفکیک نمود. به‏طور کلی نتایج مطالعه حاضر بینشی جدید در مورد پاسخ مکزیکن‌لایم به بیمارگر فیتوپلاسما طی پیشرفت بیماری فراهم نموده و در نهایت مشخص شد که می‏توان گیاهان سالم از آلوده را با بررسی روند تغییرات ترکیبات بیوشیمیایی جداسازی نمود.}, keywords_fa = {بیماری جاروک,تجزیه به مؤلفه‌های اصلی,رنگدانه,فنل,نشاسته}, url = {https://ijhs.ut.ac.ir/article_74081.html}, eprint = {https://ijhs.ut.ac.ir/article_74081_0655717389ce7a071dad5aca1aa2cfb2.pdf} }
